MONDAY, July 30 (HealthDay News) -- A new gene associated with a rare form of blindness from birth has been identified by researchers.
According to the report published online July 29 in the journal Nature Genetics, mutations in the NMNAT1 gene are linked to Leber congenital amaurosis (LCA), an inherited retinal degenerative disease that causes reduced vision in infants. Signs of vision loss are apparent in the first few months of life.
LCA is a common reason for enrolling children in schools for the blind.
This finding is a step forward in developing sight-saving gene therapy for patients with LCA, according to the researchers at the Massachusetts Eye and Ear Infirmary, the Children's Hospital of Philadelphia, Loyola University Chicago Health Sciences Division, and their colleagues.
"The immediate benefit of this discovery is that affected patients with mutations in this new LCA gene now know the cause of their condition," study co-senior author Dr. Eric Pierce, director of the Ocular Genomics Institute at Massachusetts Eye and Ear, said in an infirmary news release.
"Scientists now have another piece to the puzzle as to why some children are born with LCA and decreased vision. The long-term goal of our research is to develop therapies to limit or prevent vision loss from these disorders," Pierce said.
NMNAT1 is the 18th identified LCA gene and is located in a region known to harbor another LCA gene.
Leber congenital amaurosis occurs in about three of 100,000 newborns, and is one of the most common causes of blindness in children, according to the U.S. National Library of Medicine.

Why Mars again?
The big unknown remains. Scientists want to know if any form of life ever existed there, and that means microscopic organisms. Since the 1960s, spacecraft have zipped past, orbited or landed on Mars in this quest. Two small NASA rovers that arrived in 2004 explored different craters and one is still functioning today.
Curiosity is the most ambitious effort ever, but it's not the be-all and end-all. During its two-year exploration, it will try to answer whether the giant crater where it lands had the right conditions to support microbes. But future missions would still be needed for more answers.
What will Curiosity do?
Curiosity carries a toolbox of 10 instruments, including a rock-zapping laser and a mobile organic chemistry lab. It also has a long robotic arm that can jackhammer into rocks and soil. It will hunt for basic ingredients of life including carbon-based compounds, nitrogen, phosphorus, sulfur and oxygen, as well as minerals that might provide clues about possible energy sources.

And the International Monetary Fund warned Friday that the Spanish recession would be worse than initially expected, with a forecast contraction of 1.7 percent this year and of 1.2 percent in 2013.
Official Spanish economic growth figures for the second quarter are expected on Monday.
Spain's economy is reeling in the aftermath of a decade long real estate boom that crashed with the debt crisis, bringing the Spanish financial sector to the brink of insolvency.
Under pressure from European authorities who have agreed to bail out Spanish banks, Spain's conservative government has approved tens of billions of euros' worth of spending cuts, tax hikes and other measures.
Madrid must still pay high interest rates to borrow money on public debt markets, though the rate, or yield on benchmark 10-year bonds fell on Thursday and Friday following comments by European Central Bank chief Mario Draghi.
He declared on Thursday that the ECB was "ready to do whatever it takes to preserve the euro.
"And believe me it will be enough," Draghi added in comments that pushed yields on Italian and Spanish bonds lower because investors now expect the bank to buy debt issued by those countries, possibly in cooperation with their eurozone partners.
The stock exchange in Madrid has also leapt higher in the past two days, gaining 6.0 percent on Thursday and another 3.91 percent on Friday.
But the IMF, which wrote its report before Draghi's remarks had been made, said that heightened market tension could disrupt Spain's ability to finance itself despite a eurozone bailout worth up to 100 billion euros ($123 billion) agreed for Spanish banks and emergency financial reforms for the 17-nation bloc.
"Market tensions could intensify further, threatening market access, particularly if policies fail to stem capital outflows or due to further stress elsewhere in the euro area," the IMF said in a report.
Spanish Prime Minister Mariano Rajoy has scrambled to regain market confidence and unveiled his latest set of measures on July 11 aimed at saving 65 billion euros over three years.
Rajoy says the steps will help cut the public deficit in line with targets agreed with the European Union, and strengthen the economy in the long term.
Critics say the measures will make the poor suffer unfairly from moves such as a public sector bonus cut and a rise in sales tax that together will hit consumption.
"All the spending cut policies they are taking are restrictive and run counter to growth," said Alberto Roldan, an analyst at Spanish brokerage Inverseguros.
"Raising the fiscal pressure in a country with 25 percent unemployment is absolutely regressive."
Hundreds of thousands of Spaniards have marched across the country in recent weeks to protest against the measures.
The Spanish region hardest hit by rising unemployment was Catalonia in the northeast with a rate of 33.92 percent, Friday's figures showed. The lowest rate was 14.56 percent in the affluent Basque Country.
Concern that Spain might need a full bailout has grown since Spanish regions including Catalonia, the second biggest, said they too might need help from the federal government.
On Friday however, the government dismissed the prospect of an international rescue.
"A bailout is not an option," government spokeswoman Soraya Saenz de Santamaria said.

CORVALLIS, Ore. A new study concludes that among older adults especially those who are frail low levels of vitamin D can mean a much greater risk of death.
The randomized, nationally representative study found that older adults with low vitamin D levels had a 30 percent greater risk of death than people who had higher levels.
Overall, people who were frail had more than double the risk of death than those who were not frail. Frail adults with low levels of vitamin D tripled their risk of death over people who were not frail and who had higher levels of vitamin D.
"What this really means is that it is important to assess vitamin D levels in older adults, and especially among people who are frail," said lead author Ellen Smit of Oregon State University.
Smit said past studies have separately associated frailty and low vitamin D with a greater mortality risk, but this is the first to look at the combined effect. This study, published online in the European Journal of Clinical Nutrition, examined more than 4,300 adults older than 60 using data from the Third National Health and Nutrition Examination Survey.
"Older adults need to be screened for vitamin D," said Smit, who is a nutritional epidemiologist at OSU's College of Public Health and Human Sciences. Her research is focused on diet, metabolism, and physical activity in relation to both chronic disease and HIV infection.
"As you age, there is an increased risk of melanoma, but older adults should try and get more activity in the sunshine," she said. "Our study suggests that there is an opportunity for intervention with those who are in the pre-frail group, but could live longer, more independent lives if they get proper nutrition and exercise."
Frailty is when a person experiences a decrease in physical functioning characterized by at least three of the following five criteria: muscle weakness, slow walking, exhaustion, low physical activity, and unintentional weight loss. People are considered "pre-frail" when they have one or two of the five criteria.
Because of the cross-sectional nature of the survey, researchers could not determine if low vitamin D contributed to frailty, or whether frail people became vitamin D deficient because of health problems. However, Smit said the longitudinal analysis on death showed it may not matter which came first.
"If you have both, it may not really matter which came first because you are worse off and at greater risk of dying than other older people who are frail and who don't have low vitamin D," she said. "This is an important finding because we already know there is a biological basis for this. Vitamin D impacts muscle function and bones, so it makes sense that it plays a big role in frailty."
The study divided people into four groups. The low group had levels less than 50 nanograms per milliliter; the highest group had vitamin D of 84 or higher. In general, those who had lower vitamin D levels were more likely to be frail.
About 70 percent of Americans, and up to a billion people worldwide, have insufficient levels of vitamin D. And during the winter months in northern climates, it can be difficult to get enough just from the sun. OSU's Linus Pauling Institute recommends adults take 2,000 IU of supplemental vitamin D daily. The current federal guidelines are 600 IU for most adults, and 800 for those older than 70.
"We want the older population to be able to live as independent for as long as possible, and those who are frail have a number of health problems as they age," Smit said. "A balanced diet including good sources of vitamin D like milk and fish, and being physically active outdoors, will go a long way in helping older adults to stay independent and healthy for longer."

USC (US) ? Women who have their last baby in their 30s and 40s have a reduced risk of developing endometrial cancer, the most common gynecological cancer in the US.
New research finds that women who last give birth at age 40 or older have a 44 percent decreased risk of endometrial cancer, which strikes the tissue lining of the uterus, as compared to women who have their last baby younger than 25.
The study found that risk begins to decrease after age 30 by approximately 13 percentage points for each five-year delay in last births. Compared to women who last give birth before age 25, those who have their last child between age 30 and 34 reduce their risk by 17 percent and those between age 35 and 39 reduce their risk by 32 percent.
?While childbearing at an older age previously has been associated with a lower risk of endometrial cancer, the size of this study definitively shows that late age at last birth is a significant protective factor after taking into account other factors known to influence the disease?body weight, number of kids, and oral contraceptive use,?? says Veronia ?Wendy? Setiawan,? assistant professor of preventive medicine at the Keck School of Medicine at the University of Southern California.
Published online in the American Journal of Epidemiology, the study that is believed to be the largest of its kind, examined pooled data from four cohort studies and 13 case-control studies. Funded by the National Cancer Institute, the research examined a total of 8,671 cases of endometrial cancer and 16,562 control subjects, all derived from studies in the Epidemiology of Endometrial Cancer Consortium.
?We found that the lower risk of endometrial cancer continued for older mothers across different age-at-diagnosis groups, including under 50, 50-59, 60-69, and over 70?which shows that the protection persists for many years,? Setiawan says.
?Protection also did not vary by the two types of the disease: the more common Type 1, which we think is related to estrogen exposure; and the more rare, but more aggressive and deadly, Type 2, which has been thought to develop independent of hormones.?
Endometrial cancer is the fourth most common cancer diagnosed among American women. The American Cancer Society estimates that in 2012 about 47,130 new cases of cancer of the uterine body will be diagnosed, and about 8,010 women will die from such cancers. The vast majority of those cases are endometrial cancer.
Setiawan?s research also examines whether the association between age at last birth and endometrial cancer was consistent across race and ethnicity. The protective association was observed in Caucasian and Asian women, but not in the study?s small subset of black women, warranting additional study of larger groups of those women.
More research is necessary to determine why late age at last birth might protect against endometrial cancer, but Setiawan notes that several potential mechanisms have been suggested by other investigators, including:
Women capable of becoming pregnant at an older age may possess a healthy endometrium or experience fewer menstrual cycles without ovulation
Prolonged exposure to the hormone progesterone during pregnancy may be especially beneficial at older ages, the critical period for endometrial cancer development
Premalignant or malignant cells of the uterine cavity?s mucosal lining, which are more likely to exist with increasing age, are shed during childbirth.
?This study shows an important protective factor for endometrial cancer,? Setiawan says, ?and when the exact mechanism by which it protects women from getting the disease is known, it can help our understanding of how endometrial cancer develops and thus how to prevent it.?
